Question regarding installing a partition wall in a room with a tiled floor

I have a large 14 x 22 room. It is wall to wall ceramic tile and I want to split the room by adding a wall. Do you install the 2x4 floor footer directly on top of the tile or cut out a 3 1/2" gap in the tile where the 2x4 will then bolt to the cement slab?
 
Dealing with a similar situation in our basement right now. Previous people framed over the old laminate tiles and now I’m going to be stuck dealing with it because we would prefer to polish the concrete or do an epoxy type floor.

Cut the tile and add a baseboard with quarter round to hide the imperfections.
